Biography:

I became a Faculty Member at the Department of Biochemistry, Faculty of Basic Medical Sciences, University of Ibadan, Nigeria on the 9th of April 1998. Presently, I am a Professor of Biochemistry since October 1, 2015 and, Head of Department from August 1, 2022 till date. My research interest is in the field of Molecular Drug Metabolism and Toxicology with special interest in Drug Discovery and Development from natural products. I have studied the antioxidative and free radical scavenging effects of some indigenous phytochemicals (Artocarpus altilis, Curcuma longa, Cocos nucifera, Garcinia kola, Xylopia aethiopica, Calliandra portoricensis, etc) in both in vivo and in vitro experiments. In functional experiments, l have studied the underlying mechanism of cardiovascular effects of kolaviron, a biflavonoid complex from Garcinia kola. For the first time, l have shown that kolaviron acts directly on ion channels in vascular beds via blockage of extracellular Ca2+ influx, inhibition of intracellular Ca2+ release and opening of K+ channels that are sensitive to 4-aminopyridine and charybdotoxin. Also, I have worked on the anticancer and anti-angiogenic potentials of Xylopia aethiopica, Calliandra portoricensis, punicalagin, methyl jasmonate, etc in C-33A, MCF-7, PC-3 and LNCaP cancer cells. Presently, l have developed an experimental model of benign prostatic hyperplasia (BPH) in castrated rats. In my laboratory, we have started screening array of bioactive compounds as anti-BPH agents. We have shown for the first time that methyl jasmonate and finasteride synergistically protect against BPH in rats via mechanisms that involve anti-inflammation, induction of apoptosis and inhibition of phase I drug metabolizing enzyme. Also, l have developed experimental animal model of metastatic mammary tumour in female Wistar rats. In my Lab, we have shown that fractions from Calliandra portoricensis, Annona muricata and kolaviron (biflavonoid fraction from Garcinia kola) attenuated inflammation, induced apoptosis and restored cyto-architecture of mammary gland in rats administered N-methyl-N-nitrosourea and benzo(a)pyrene. Research is ongoing to evaluate imparts of fractions from phytochemicals on metastatic cancer cells into distant organs in the malignant rats.

Title: Ameliorative effects of fractions from Calliandra portoricensis on the development of hepatocellular carcinoma (HCC) in metastatic mammary tumour-induced by N-methyl-N-nitrosourea and benzo(a)pyrene in Wistar rats

Brief Summary: The study aims to evaluate in a mechanistic term, the role of fractions from Calliandra portoricensis in inhibiting the migration of metastatic cancer cells from cancerous mammary gland to hepatocytes of the animals. Biochemical assays which include Chick Chorioallantoic Membrane assay (angiogenesis), Migration assay (metastasis), NF-kB and IL-1β assays (inflammation), histology of hepatocytes (direct visualization of cancer cells), survival assay, etc will be conducted in this study.

Completed Research

The adverse effects of antihypertensive drugs, such as Ca2+ channel inhibitors, alpha and beta blockers, etc are well documented. The need to protect humans against hypertension and cardiovascular diseases (CVD) using newer and safer class of antihypertensive drugs is important. Therefore, a search of natural products that can protect humans from CVD is a key issue in cardiovascular research. The use of plant products may be beneficial in protecting against CVD since they are less toxic than the synthetic compounds. Recently, soluble epoxide hydrolase (sEH) inhibitors are gaining attention as potent antihypertensive agent. This enzyme converts epoxyeicosatrienoic acids (EETs), a metabolite from arachidonic acid to dihydroxyeicosatrienoic acids. This reaction results in a loss of biological activities of EETs, in particular its ability to mediate in vasodilation as endothelium-derived hyperpolarizing factors. Therefore, agents that can inhibit sEH synthesis will elicit antihypertensive effect. We screened crude extracts from Garcinia kola, Alchornea laxiflora, Alstonia boonei, Morinda lucida, Curcuma longa L, Xylopia aethiopica, Mallotus oppositifolius, etc, as putative inhibitors of sEH. The in vitro high throughput screening of these extracts was achieved using HPLC method. Furthermore, functional studies involving calcium imaging with fluo-4-AM dye were conducted to ascertain the effect of extracts on calcium signals in transfected HEK 293 cells capable of expressing TRPV1 585i ion channel.
